Development and use of an internal positive control for detection of Bordetella pertussis by PCR.
An internal control of amplification was constructed by recombinant PCR to detect PCR inhibitors. This exogenous DNA was included in the reaction mixture and coamplified with the target gene. This detection was successfully applied to the diagnosis of whooping cough by amplification of a fragment of Bordetella pertussis IS481.
متن کاملEpidemiological consequences of an ineffective Bordetella pertussis vaccine
The recent increase in Bordetella pertussis incidence (whooping cough) presents a challenge to global health. Recent studies have called into question the effectiveness of acellular B. pertussis vaccination in reducing transmission. Here we examine the epidemiological consequences of an ineffective B. pertussis vaccine. Using a dynamic transmission model, we find that: 1) an ineffective vaccine...

Abstract False positive results are the major problem influencing interpretation of Clinical Laboratory test. They are originated mostly in the other diseases, technical errors and the recent vaccination. The problem has been presented since 1991 is positive HIV test after influenza Vaccination (1). The False positive has been reported in Cases using Eliza, one of The most common test to screen...
متن کاملReal-time PCR-based detection of Bordetella pertussis and Bordetella parapertussis in an Irish paediatric population.
Novel real-time PCR assays targeting the Bordetella pertussis insertion sequence IS481, the toxin promoter region and Bordetella parapertussis insertion sequence IS1001 were designed. PCR assays were capable of detecting ≤10 copies of target DNA per reaction, with an amplification efficiency of ≥90 %. From September 2003 to December 2009, per-nasal swabs and nasopharyngeal aspirates submitted f...
متن کاملUnexpected isolation of Bordetella pertussis from a blood culture.
Bordetella pertussis was isolated from a culture of blood from a 31-year-old man with Wegener's granulomatosis. The organism was detected with the BACTEC 9240 system after 6 days of incubation and was confirmed as B. pertussis by the Centers for Disease Control and Prevention. To our knowledge, this is the first published report of the recovery of B. pertussis from blood.
